Simmons gained 161 yards and scored three times, while Lindsay ran for 158 yards - 118 in the first half - scored once and threw a 65-yard touchdown pass.
The Ragin' Cajuns (1-9) provided the perfect tonic for an Oklahoma State team (4-6) that has been frustrated by near-misses this season.
The Cowboys, who saw their chances for a winning season dissipate with a loss to Texas last week, had 224 yards in the first quarter and finished with a season-high 605. Their 440 rushing yards were the most since gaining 536 against Kansas in 1988.
Lindsay set the tone by gaining 11 yards on a keeper on the Cowboys' first play. Oklahoma State drove 81 yards on just five plays, with Simmons scoring on a 21-yard run.
Lindsay's 56-yard keeper down the right sideline set up his 1-yard touchdown late in the first quarter, making the score 17-0. He had a 22-yard run in the second quarter that helped lead to Simmons' second touchdown, a 1-yard run.
Lindsay also threw a 65-yard touchdown pass to Ethan Howell early in the third quarter. It came on the first snap after Howell's twin, Evan, intercepted Dere Dyer for the second time.
About the only mistake Lindsay made came early in the second quarter when he threw a pass that was intercepted by linebacker Matt Cieslak and returned 18 yards for a touchdown.
Simmons' final score came on a 20-yard run with 5:41 left in the third quarter, making it 44-6. He and Lindsay watched from that point on.
Southwestern Louisiana totaled 241 yards, but 150 came in the fourth quarter when the Ragin' Cajuns scored on passes of 16 and 61 yards from Lance Domec to Brandon Stokley.
Dyer had a miserable afternoon, completing just four of 14 passes for 43 yards and three interceptions. He also was sacked five times.
